Gateway enzyme for chemicals from catnip to cancer drug discovered

ScienceDaily (Nov. 21, 2012) ? Scientists have discovered an enzyme used in nature to make powerful chemicals from catnip to a cancer drug, vinblastine. The discovery opens up the prospect of producing these chemicals cheaply and efficiently.

They are produced naturally by some plants such as the medicinal Madagascar periwinkle, but faster-growing plants could be used to produce them. With synthetic biology, improvements could also be made to them.

The study, to be published in Nature on November 22, was led by scientists from the John Innes Centre, an institute on Norwich Research Park strategically funded by BBSRC.

"Thousands of chemicals are derived from the enzyme we have called iridoid synthase," says senior author Dr Sarah O'Connor from the JIC and the University of East Anglia.

"We can start to use it to come up with new-to-nature structures with biological activity of benefit to both medicine and agriculture."

Many aphids, often important agricultural pests, produce sex pheromone chemicals that are identical to or that closely resemble the iridoid synthase product. Strategic use of these iridoid chemicals could be used to disrupt the aphids' breeding cycle or to repel them from crops.

The anticancer ingredient vinblastine sulphate is currently derived from the Madagascar periwinkle plant. The iridoid synthase is an essential step in the production of this compound. But vinblastine is produced in just very low levels and the drug has many side effects. The hope is to find a way to produce it more cheaply, easily and with a chemical structure that lessens side effects.

"We need to identify more enzymes to see the entire pathway used in nature to make this potent compound," said Dr O'Connor.

"But the enzyme we have discovered is also the basic scaffold for many other iridoid chemicals and we can start to experiment with building new chemical structures with biological activity."

The backbone of all iridoids consists of two fused rings and scientists have been trying to track down what makes this ring system. Experiments showed that iridoid synthase is the enzyme responsible.

Scientists already knew the enzyme preceding iridoid synthase and how the gene encoding it is expressed. The lead author, Dr Fernando Geu-Flores from JIC, therefore looked for enzymes that are encoded by genes expressed in a similar way and narrowed down their search to 20 enzymes.

Research published in the 1980s indicated that the missing enzyme is dependent on a particular compound called NADPH, which narrowed the search down to two enzymes.

O'Connor and her co-workers will also investigate whether the enzyme is important in a simple chemical reaction used by chemists for nearly 100 years. Scientists are trying to identify which enzymes catalyse a reaction called the Diels-Alder reaction, named after the Nobel prize-winning scientists who discovered it. A better understanding of how the iridoid synthase works could open up new ways to make pharmaceutical compounds using synthetic biology.

M23 rebels vow to take Congo, as troops defect

GOMA, Congo (AP) ? Pressing ahead with their seizure of cities in mineral-rich eastern Congo, the M23 rebels said Wednesday they are fighting to control all of this sprawling country and to topple President Joseph Kabila's government.

Following their capture of the strategic city of Goma a day earlier, the rebels took the nearby town of Sake on Wednesday as they moved toward the provincial capital of Bukavu.

"Kabila has to go. We want our country back," said M23 Col. Vianney Kazarama to cheers from thousands gathered at the Goma stadium. "We are now going to Kinshasa. No one will divide this country."

Nearly 3,000 Congolese army soldiers and police defected to the rebels in Goma on Wednesday and turned in their weapons at the stadium rally.

Even as the M23 rebels consolidated their gains, the presidents of Congo, Rwanda and Uganda called on them to give up the territory they now control.

The M23 is made up of hundreds of officers who deserted the Congo army in April this year. Neighboring Rwanda has supplied soldiers and equipped the rebels with sophisticated arms, including night vision goggles and 120 mm mortars, according to a report by United Nations experts to be published Friday.

The U.N. report accuses the rebels of grave human rights abuses included murder, rape and the use of child soldiers.

The M23 is largely made up of ethnic Tutsis who want to extend their control over eastern Congo and its valuable deposits of gold, copper, coltan and timber. Their campaign, and alleged support from Rwanda, has its roots in the 1994 genocide in which some 800,000 Tutsis were massacred by Hutus.

Since then eastern Congo has been plundered by several rebel forces and foreign armies in conflicts that killed millions of people.

The speed with which the M23 has seized Goma and the surrounding countryside exposes the weakness of Kabila's government, 1,500 kilometers (978 miles) to the west in Kinshasa, experts said.

"It's a setback for Kabila. He wasn't even capable of reforming the army and the fall of Goma demonstrated that," said Felix Bafilemba, a Goma-based analyst for the Enough Project. "It marks the beginning of the end for Kabila's regime."

Strewn with bodies, burned out vehicles and abandoned tanks, the road between Goma and Sake showed evidence of Tuesday's fighting.

M23 soldiers in vehicles patrolled the streets of Sake, 27 kilometers (17 miles) south of Goma. As in Goma, most residents accepted the rebels' presence.

"We only want peace," said Innocent Rumabo in Sake.

Other resident complained of rapes carried out Tuesday night.

"I know two women who were raped, there are more in the area," said one Goma resident who insisted on anonymity for fear of retaliation.

In neighboring Uganda, the presidents of Uganda, Rwanda and Congo resolved to put pressure on the M23 rebels to give up territory they have captured, according to a statement released at the end of the emergency talks.

"Even if there are legitimate grievances by the mutineering group known as M23, they cannot accept the expansion of this war or entertain the idea of overthrowing the legitimate government of (Congo) or undermining its authority," said presidents Joseph Kabila of Congo, Paul Kagame of Rwanda and Yoweri Museveni of Uganda, in a statement issued late Wednesday.

"Therefore the M23 must immediately stop this offensive and pull out of Goma," said the presidents' statement issued in Kampala. This resolution was to be immediately communicated to the M23 leadership, the statement said.

Behind the statement, there was considerable bargaining in the emergency summit, according to diplomats. Congo's Kabila made a concession by agreeing to negotiate directly with the M23, a reversal from his previous refusal to talk with the rebels.

For his part, Kagame publicly supported Kabila and reiterated his denial that Rwanda is supporting the rebels. But U.N. experts say the evidence is overwhelming that Kagame is supporting, even directing, the M23 so that tiny Rwanda can control eastern Congo's lucrative mineral trade.

Uganda, which was also implicated in the Congo violence by report of U.N. experts, denies supporting the rebels but Museveni is increasingly seen as an ally of Rwanda's Kagame.

While the three presidents called on the M23 to give up the territory that they have seized, the rebels continued Wednesday on toward their next target: Bukavu, the capital of South Kivu province on Lake Kivu.

The Congo soldiers remaining in the government army near Goma said they are not sure what to do.

"We are waiting for orders now. We don't know what we are supposed to do. It's hard. My comrades who defected in Goma, we're going to fight them," said a Congolese army major reached by phone in Kanyabayonga, northwest of Goma, where some of the troops have regrouped. The major required anonymity because of the uncertain situation.

The U.N. has its largest peacekeeping force in the world in Congo, with 18,000 troops, known by their acronym MONUSCO. But the 1,500 peacekeepers in Goma did not help the Congo government troops forces during Tuesday's battle because they do not have a mandate to engage the rebels, said Congolese military spokesman Olivier Hamuli, who expressed frustration over the lack of action by the peacekeepers.

A U.N. spokesman in New York said the peacekeepers in Goma held their fire to avoid triggering a battle.

On Wednesday, the Security Council was expected to review the mandate of the U.N. peacekeeping force in Congo. A resolution adopted Tuesday by the Security Council asks the U.N. secretary-general to recommend possible redeployment, and possible "additional force multipliers."

The resolution approved unanimously by the council imposes targeted sanctions, including a travel ban and assets freeze on the M23 rebel group leadership. But it did not name two countries accused by Congo of supporting the rebels: Rwanda and Uganda.

The council demanded that the M23 rebels withdraw from Goma, disarm and disband, and insisted on the restoration of the crumbing Congolese government authority in the country's turbulent east.

Migration as a cross-cultural marital metaphor? On structuring a ...

Yesterday, I explained that I had started writing this memoir by using the metaphor of driving ? as I think of the marriage as a road trip. ?However, now that I think about it, although I identified that as a red thread yesterday, it actually may be more of a structure, such as the one my friend Sinan spoke of. So, whether it is a red thread or a structure or whatever it may be ? let tell you how I got to this place (?as if there were a choice!,? Karag?z, the sarcastic trickster puppet?points out.) Reflecting further, I thought, we do travel between worlds both while in Turkey and while in the United States. ?M. always says he feels more American in Turkey now. ?I am often thinking about Turkey while engaging in normative American behaviors ? or Turkish-American behaviors. ?We often talk about Turkey. ?We eat Turkish food every day. ?We drink Turkish tea all the time ? and on and on. ?And given that idea, that we travel between worlds on a constant basis at both the?micro and a macro levels, what does a conceptual framework for migration?do for structuring this memoir ? or for finding my red thread?

So the conceptual model of migration I found goes like this: ?When working with immigrants to a new country, there are three phases they may be in. ?And, as a social worker, one should assess for which phase that person is in so that you can put yourself in their shoes some. ?Those three phases are:?Pre-migration;?Transition and Re-settlement. ?Now, as a student of the impact of globalization and transnational migration patterns, I know full well that this conceptual model is flawed ? as many people only migrate on a temporary basis, with the intention of returning ?home? someday. ?With globalization we are exposed to cultures and subcultures constantly, able to rotate between places and homes and cultures very easily, if the penny permits, we are not isolated ? and we are dealing with it all, all at once. So, this started to make sense vis-a-vis how I experience my cross-cultural marriage. ?There was something there I felt I could work with.

So, I began to think about migration. Now, I have not migrated anywhere permanently, except perhaps in my own mind. ?And I am hardly an expatriate at this point, although that is the group of folks I feel most akin to. ?And that is why the workshops conducted by those super-smart and interesting ladies over at Global Niche are calling me like a siren song ? as they can relate to the confusion of place, culture and identity in the cross-hairs of in-between. M. is with me in those cross-hairs, and?I think that somehow our time both in the United States and in our extended stays in Turkey has caused me to migrate my mind and perhaps my thinking and some day-to-day practices of life. As for M., he migrated from Turkey to the United States in part as a way out of tradition and customs that did not feel right to him, akin to rubbing a dog?s hair the wrong way, constantly. ?And perhaps that is why life in another culture comes easily to him, I don?t know. Migration is also a comfortable concept to me ? my Grandmother and Grandfather emigrated to New York City from Spain and Scotland, respectively, and never quite found themselves here or there. ?My other Grandmother emigrated from Quebec province as a baby, and the spectre of other-ness haunted her for a time. ?My stepmother and stepsiblings grew up in Kenya and East Pakistan (now Bangladesh) and migrated an international fiber into the fabric of our blende family. My good friend Trisha Thomas is a cross-cultural mother ? an American living in Italy for many years now ? and she thinks on these issues all the time over at her blog, Mozzarella Mamma

So, yes, I grew up with a well-steeped and stewed sense of other-ness, and mixed-ness vis-a-vis my family?s cultural elements and the migration patterns that led to that. ?And as a result of all that,?here is the structure I have today ? option #2 ? and who knows where it will go from here??

Section 1: Pre-migration:?This section of the book will be a series of chapters about my early exposure to Islam, Muslims and countries in the Middle East. ?It would also address my youthful obsession with being different ? and exploring different cultures. ?It will end with a chapter on meeting M. and the beginnings of my foray into a cross-cultural relationship.

Nordeus partnership for bwin.party...

The world?s largest listed online gambling operator, bwin.party digital entertainment, has signed a deal that will see it develop a social sportsbetting application in partnership with Serbian firm Nordeus.

Following the launch of its bespoke Win business unit in May, bwin.party stated that the agreement is a ?further step forward? in the execution of its ?social gaming strategy? as it looks to capitalise on its ?strong presence in real-money sportsbetting?.

(AP) ? A measure of the U.S. economy intended to signal future activity rose only slightly last month, suggesting growth could stay weak.

The Conference Board said Wednesday that its index of leading indicators increased 0.2 percent in October after a 0.5 percent gain in September. The index is intended to anticipate economic conditions three to six months out.

The strength in October came from lower interest rates, a drop in applications for unemployment benefits, and an increase in demand for large manufactured goods.

Applications for unemployment aid have spiked this month because Superstorm Sandy closed businesses and cut off power to 8 million homes in 10 states. People can claim unemployment benefits if their workplaces are forced to close and they aren't paid.

In all, only four of the 10 indicators that make up the index improved. The major areas of weakness were a decline in building permits, a drop in consumer confidence, a decline in the Institute for Supply Management's new orders index and lower stock prices.

"Based on current trends, the economy will continue to expand modestly through the early months of 2013," Ken Goldstein, an economist at the Conference Board, said.

Many analysts predict growth at an annual rate of roughly 3 percent in the July-September quarter, up from the initial estimate of 2 percent reported last month. The government releases its second estimate for third-quarter growth on Nov. 29.

But many economists expect growth to slow in the current October-December quarter to an annual rate below 2 percent. Part of the slowdown is likely to occur because of Superstorm Sandy, which disrupted businesses and cut off power throughout the Northeast.

Many companies may also hold off on hiring and new investment until the White House and Congress resolve the "fiscal cliff," the package of tax cuts and spending increases scheduled to take effect Jan. 1.

If the cliff is avoided, many analysts think growth could accelerate next year.
